Effect of Soybean (glycine max) Leaf Extract against the Development of Non-alcoholic Fatty Liver in High Fat Diet-fed Mice

Abstract
This study was designed to investigate the effect of soybean leaf extract on the development of hepatic steatosis in high fat diet-fed mice. Soybean plants were treated with ethephon to enhance the production of isoflavones in the leaves. The mice were fed with normal chow diet(ND), high fat diet(HFD) or HFD+Soybean leaf extract for 9 weeks. HFD feeding significantly increased the body weight gain and visceral and subcutaneous adipose tissue weights compared to ND group in the mice, but these increases were attenuated by Soybean leaf treatment. Soybean leaf also attenuated the increases in hepatic lipid droplet and hepatocellular triglyceride level at 64 days of HFD feeding. Finally, soybean leaf markedly increased the phosphorylation of AMPK and ACC and their protein expression in the liver at 64 days of HFD feeding, while inhibited the increase in fatty acid synthase protein expression. Our results indicate that soybean leaf extract has beneficial effects against hepatic steatosis in HFD-fed mice.
